Leptin is a peptide hormone that plays a very important role in the regulation of energy intake and expenditure in the human body. It affects appetite and metabolism. The discovery of leptin in 1994 was thanks to a study by researchers on the causes of extreme obesity in a certain type of mouse. The researchers concluded that they had a mutation in the gene responsible for leptin synthesis.
As leptin suppresses appetite strongly, its absence in mutated mice led them to uncontrollable eating, which also caused obesity.
The leptin gene is also called the obese gene and is located on chromosome seven. The main part of leptin that circulates in the human body is produced by adipose tissue and only minimal amounts are secreted by the epithelial cells of the stomach and placenta.
Once in the blood, leptin is transported to the hypothalamus in the brain, where it stimulates the center of satiety and pleasure. The amount of leptin in the body increases with the increase in body fat.
